Spider neurotoxins and their neuronal receptors

چکیده

Seven high molecular weight neurotoxins which stimulate secretion of various neurotransmitters were isolated from the venom of black widow spider Latrodectus mactans tredecimauttatus. Among them a-latrotoxin is very specific for vertebrate while latroinsectotoxins are active only for insects, a-latrocrustatoxin affects crustacean nerve endings. Latrotoxin preparation consists of 130 and 8 kD proteins. The cDNAs encoding a-latrotoxin and a-latroinsectotoxin were cloned and sequenced. Membrane receptors for all neurotoxins were studied in nerve tissue preparations of appropriate animals. a-Latrotoxin was shown to open the ion channels in the membrane of XenoDus oocytes injected by brain mRNA fraction.

alpha-Latrotoxin and its receptors: neurexins and CIRL/latrophilins.

alpha-Latrotoxin, a potent neurotoxin from black widow spider venom, triggers synaptic vesicle exocytosis from presynaptic nerve terminals. alpha-Latrotoxin is a large protein toxin (120 kDa) that contains 22 ankyrin repeats. In stimulating exocytosis, alpha-latrotoxin binds to two distinct families of neuronal cell-surface receptors, neurexins and CLs (Cirl/latrophilins), which probably have a...

The Role of Adrenergic Receptors on Neural Excitability and Synaptic Plasticity: A Narrative Review

Adrenergic receptors have an important role in neural excitability and synaptic plasticity. Despite a lot of studies on these receptors, their exact role in brain disorders accompanied with hyperexcitability has not been determined. There are also controversies on their role in synaptic plasticity. In this review article, the important studies done in this regard have been reviewed to achieve a...

Neurotoxins from Wecffewys Spider Venom are Potent Presynaptic Blockers in Drosophila

Studies of presynaptic events in synaptic transmission may be facilitated through the use of specific ligands for functional components of the transmitter release mechanism and through the use of genetics. For this purpose, neurotoxins that affect neuromuscular transmission in Drosophila have been identified and purified from Plecfreurys spider venom (PLTX). One class of toxins causes irreversi...

Snake venom a-neurotoxins and other `three-finger' proteins

The review is mainly devoted to snake venom a-neurotoxins which target different muscle-type and neuronal nicotinic acetylcholine receptors. The primary and spatial structures of other snake venom proteins as well as mammalian proteins of the Ly-6 family, which structurally resemble the `three-finger' snake proteins, are also briefly discussed. The main emphasis is placed on recent data charact...

Structural and functional characterization of a novel homodimeric three-finger neurotoxin from the venom of Ophiophagus hannah (king cobra).

Snake venoms are a mixture of pharmacologically active proteins and polypeptides that have led to the development of molecular probes and therapeutic agents. Here, we describe the structural and functional characterization of a novel neurotoxin, haditoxin, from the venom of Ophiophagus hannah (King cobra). Haditoxin exhibited novel pharmacology with antagonism toward muscle (alphabetagammadelta...
